Abstract

The periapical lesion is an inflammatory response to infection in the root canal system.roots, with the participation of different cell types, receptors and ligands during theits development. Pathogen-associated molecular patterns (PAMPs) as well assynthetic ligands such as Pam3Cys, bind to toll-like receptors, such as TLR2,to activate the innate immune system. Regulatory T lymphocytes (Tregs) are important cellsin the defense system, as well as neutrophils and their extracellular traps (NETs). In additionIn addition, chemokines such as CXCL5 are present to carry out chemotaxis. the purpose of thisstudy is to evaluate the role of Tregs, NETs and CXCL5 in the presence or absence of Pam3Cys inperiapical lesions induced in wild-type (WT) mice and tolllike receptor 2 knockout (TLR2KO). In the study, samples from another study already carried out will be used. they wereWT and TLR2KO mice were used, allocated in control groups (healthy molars) andexperimental (induction of periapical lesion), subdivided into 3 periods (14, 21 and 42 days). to definedays after induction, Pam3CSK4 was injected intraperitoneally in some groups. After this,the animals were euthanized, the mandibles dissected and demineralized tohistotechnical processing. The immunohistochemical reactions will be performed using theFOXP3 markers for labeling Tregs, neutrophil elastase with histone H3citrullinated and meloxyperoxidase with citrullinated histone H3 for visualization of NETs. For yourtime, a CXCL5 will be labeled by the CXCL5 antibody. The results will be expressed from thetick count. The data will be analyzed using a statistical program,using the appropriate tests for each analysis, with a significance level of 5%.
