Abstract

The identification of the endothelium as a major source for catecholamines, and their role as main modulators of the cardiovascular system, has provoked a change in our understanding of the physiology and pharmacology of the circulation. 6-Nitrodopamine (6-ND) has been described as the most potent endogenous positive chronotropic and inotropic agent ever described, with different mechanisms of action of the classical catecholamines. We have now preliminary results that both rabbit and swine isolated bronchi and lung parenchyma do present basal release of 6-ND. Considering that 6-ND in the vasculature acts as a very potent vasorelaxant, the potential of this catecholamine and/or other nitro-catecholamines (6-nitrodopa and 6-nitroadrenaline) or cyano-catecholamines (6-cyanodopamine) to modulate bronchial smooth muscle tonus is of great therapeutic importance. Thus, this project is concerned with the evaluation of the role of 6-ND (and eventually other catecholamines mentioned above) in the respiratory system, employing both in vitro and in vivo models.
