Effect of administering 325 mg of recombinant bovine somatotropin (Posilac®) on the oocyte quality of Holstein heifers subjected to follicular aspiration and in vitro embryo production

Abstract

The aim of this project is to evaluate the effects of administering 325 mg of recombinant bovine somatotropin (bST; Posilac®) on the quality and quantity of oocyte cumulus complex (COCs) recovered by follicular aspiration and on the efficiency of in vitro embryo production (IVEP) in Holstein heifers. The central hypothesis is that the treatment with bST at the beginning of follicular growth wave synchronization (D0), seven days before follicular aspiration (D7), will result in greater quantity and better quality of recovered COCs, improving efficiency in IVEP and resulting in higher pregnancy rate by embryo transfer (ET). For this, 80 Holstein heifers with similar body condition score, age and weight will be used. On D0, 2 mg of estradiol benzoate, 175 mg of injectable progesterone and 0.526 mg of sodium cloprostenol will be administered. On D7, follicular aspiration will be conducted for IVEP and subsequent ET. The females will be allocated into two experimental groups on D0, bST group (n=40) will be treated with 325 mg of bST and the control group (n=40) will receive no additional treatment. The quantity and quality of viable COCs, cleaved structures, blastocysts produced and the pregnancy rate of the recipients will be the variables determined. The data will be analyzed using the MIXED, FREQ and GLIMMIX procedures in SAS(SAS Institute Inc. Cary, NC, EUA; versão 9.3).
