Abstract

Physical inactivity and sedentary behavior are associated with increased risk of chronic diseases and all-cause mortality. Acute interruptions or reductions in prolonged sedentary time seem to offset markers of cardiometabolic disturbances. However, the effects on lipid metabolism, and how long these benefits are sustained remain unknown. The aim of this research proposal is to investigate the short-term (i.e., 6 weeks) effects of brief active breaks in sedentary behaviors compared to continuous aerobic exercise on oxidation and traffic of lipids. Inclusion criteria include: i) adults with overweight/obesity aged 19-45 years; ii) being physically inactive (< 150min/week moderate to vigorous physical activity); and sedentary (> 6 h/day sitting). Participants were randomly assigned to brief active breaks intervention (BREAK) or continuous aerobic exercise (ONE). Participants in the BREAK group underwent 6 weeks of brief active breaks in prolonged sitting (i.e., 9 hourly 5-min breaks over 9 consecutive hours, 5 days/week), while participants in the ONE group performed a single, continuous bout of brisk walking (i.e., 45-min/day, 5 days/week). Oxidation and traffic of lipids and carbohydrates, total energy expenditure and fluctuation use of energy substrates were assessed by a 23h-period in a whole-room calorimetry. Participants ingested a standardized liquid shake with addition of U13 glucose and D31 palmitic acid for analysis of carbohydrate and lipid oxidation, respectively. The innovative aspects of this research proposal include: (i) to investigate whether nutrient oxidation responses observed in short-term interventions and controlled environment are sustained with longer and real-world interventions; (ii) to elucidate the physiological mechanisms underlying nutrients oxidation after the interventions. In addition, this research internship will provide an outstanding professional and personal experience to the candidate and will contribute for the development of new and productive research partnerships.
