POROUS IMPLANTS ASSOCIATED TO TERIPARATIDE DRUG DELIVERY: IMPACT ON PERIMPLANTAR REPAIR OF ESTROGEN DEFICIENCY RATS

Abstract

Osteoporosis is a metabolic disease that involves a decrease in bone quality and density, conse-quently reducing bone strength. This disease affects many women in the postmenopausal period due to the lack of estrogen supply after the reproductive stage. This deterioration in bone quality and architecture interferes with the primary and secondary stabilization of the implant, requiring ways to overcome this impasse. As alternatives to improve osseointegration, macro-geometric changes in the implant and local drug delivery stand out, which are responsible for increasing the contact between bone and implant, promoting cell growth and differentiation, and assisting in new bone formation. Currently, the most prescribed drugs for osteoporosis treatment belong to the bisphosphonate class; however, situations such as osteonecrosis and decreased cell turnover make them less attractive compared to anabolic drugs such as teriparatide, which stimulates bone neoformation and increases the osteoblastic population, besides being used as therapy for mandi-bular/maxillary osteonecrosis. Thus, the present study aims to analyze the performance of im-plants with biomimetic changes associated with drug delivery during the bone repair process. For this, 4x rats will be divided into the following groups: G1 SF (solid implant without functionali-zation), G2 SF (porous implant without functionalization), G1 PTH (solid implant with teripara-tide), and G2 PTH (porous implant with teriparatide). The rats will undergo ovariectomy surgeri-es and implant installation, followed by euthanasia for sample collection destined for histological, computed microtomography, and confocal microscopy analyses.
