Abstract

Head and neck radiotherapy (RT) causes damage to adjacent healthy tissues and leads to changes in the local microbiota, as well as alterations in host defense processes. The objective of this clinical study is to evaluate the microbialprofile and the difference in the periapical inflammatory response of single-rootedteeth with primary endodontic infection (PEI) and periapical lesion (PL), in patients who underwent RT or not. Twenty-four teeth will be selected and divided into 2 groups: 1) patients who underwent RT in the last 6 months (n=12); 2)patients who did not undergo RT (n=12). To evaluate the bacterial load, sampleswill be collected from the interior of the canals immediately after coronal opening (S1) and after biomechanical preparation (S2) for subsequent analysis by the qPCR method. To quantify the inflammatory cytokines TNF¿, IL-1¿, IL-6, IL-10,and IL-17, collections of the periapical interstitial fluid will be performed afterbiomechanical preparation (SF) and will be evaluated by the multiplex assay. Thequantified data will be analyzed and compared using appropriate statistical tests(p<0.05).
