Abstract

In this research project, we aim to apply the innovative and promising strategy of fragment-based drug discovery (FBDD), an approach that employs small molecules, known as fragments, to identify and optimize interactions with therapeutic targets. The primary focus will be on the fragment 5,8-dihydro-1,6-naphthyridin-7(6H)-one, aligned with the advancements of the team at Durham University, identified through their heterocyclic fragment library (ROF2). The synthesis of this fragment will be conducted using an already validated methodology, with the potential for optimization to enhance efficiency and reduce the complexity of the reaction steps. Vectorial functionalization will be explored through metalation and radical reactions, alongside the integration of enabling technologies such as continuous flow, photocatalysis, and electrochemistry, which may expand the diversity of the methodologies. Additionally, we aim to consolidate our research group's expertise in the functionalization of heteroaromatics, emphasizing the generation of novel drug candidates by leveraging the innovative methodologies and advanced technologies developed over the years.
