Abstract

Amorphous solid dispersions (ASDs) have proven to be an effective strategy to address the low solubility of many drugs, a common challenge in pharmaceutical development. This project proposes a comparative analysis between the extended Flory-Huggins (xFH) and PC-SAFT models for predicting miscibility and solubility in ASDs. The xFH model incorporates compositional dependence into the drug-polymer interaction parameter, enabling greater accuracy at low drug fractions, while PC-SAFT employs a more detailed thermodynamic approach. Validation will be conducted using experimental data obtained through differential scanning calorimetry (DSC) and solubility tests and literature data for various drug-polymer combinations. By comparing the precision and applicability of both models, this project will contribute to improving ASD design, enhancing the stability and efficacy of pharmaceutical formulations.
