Abstract

Single nucleotide polymorphisms (SNPs) in genes related to magnesium (Mg) metabolism have been associated with metabolic syndrome. Mg acts as a cofactor in more than 300 enzymatic reactions, including kinases, cellular metabolism, protein synthesis, and DNA. SNP analysis alone may not be sufficient to assess the risk of diseases or nutritional deficiencies, and thus the construction of a polygenic risk score (PRS) offers an option to assess the interaction of several SNPs in this context. This research aims to determine whether a PRS is associated with biomarkers of metabolic syndrome and Mg status in adults and elderly participants in a population-based study in São Paulo. This cross-sectional study includes 294 adult and elderly participants from the second phase of ISA-Nutrição 2015. A PRS will be developed for each group based on 10 SNPs related to Mg metabolism and metabolic syndrome. Dietary intake and serum Mg concentration will be evaluated as biomarkers, establishing a cutoff point for serum Mg in each group. Statistical analyses will include mean tests, correlations and multivariate regression models, with a significance level of 5% (p<0.05).
