Abstract

The giant cell tumor of bone (GCT) is a destructive osteolytic neoplasm that represents 5% of all the primary bone tumor, although benign presents a aggressive local behavior. The tumor shows specific histological features, being composed by mononuclear cells and multinuclear giant cells with variable potential for growth. Even the GCT with benign characteristics can evolve with local metastases, pulmonary metastases, in lymph nodes and more rarely malignant transformation. Studies involving analysis of gene expression has identified several genes whose expression is reduced in some types of tumors, as carcinoma of head and neck and clear cell renal carcinoma. Many molecular mechanisms may be involved in the differential expression of these genes in tumor tissue. This project has the aim of evaluating the methylation profile of the genes DUSP1, CSTB, GPC3, KTN1 and ZAK showing low expression in GCT and some other tumor types already described in literature, through MSP technique, which offers a qualitative assessment of the methylation state of CpGs islands. Thus, the assessment of the methylation profiles of selected genes may contribute to the development of more effective diagnosis and prognosis for giant cell tumor, and for developing new anti-neoplastic drugs.
