Influence of doxycycline and minocycline on ligature-induced periodontal disease in rats: a histological, histometric and histochemical evaluation

Abstract

Periodontal disease (PD) has a multifactorial etiology, which involves local aspects and systemic factors. The tissue degradation observed in PD is due to the bacterial-endotoxins exposition associated to immuno-inflammatory response of the host. The use of immuno-modulatory agents associated with conventional periodontal therapy intends to produce better results during PD treatment. Tetracyclines used in sub-clinical doses are able to modulate the immuno-inflammatory response by different pathways. The aim of the present research will be to evaluate the effect of two tetracyclines (doxycyxline – Dx and minocycline – Mn) on PD ligature-induced in rats. Forty male rats will be divided into four groups: G1 (n=10) – without ligature and drugs; G2 (n=10) – with ligature and no drugs; G3 (n=10) – with ligature + Dx; G4 (n=10) with ligature + Mn. The evaluation period will be 30 days. The following parameters will be observed: a) histological and histometrical evaluation – the junctional epithelium, connective tissue, root cement, collagen and elastic fibers and bone loss (HE and Gomori Trichromic coloration) and; b) histochemical analysis - osteoclasts activity through tartarate resistant acid phosphatase enzyme (TRAP). Data analysis will be performed by parametric and non-parametric models. From the results of the present study, we hope to obtain a better comprehension over the different mechanisms of PD inflammatory modulation by tetracyclines. (AU)
