Abstract

Gums and polymers are some names for materials that are capable of making water systems more thick and viscous or form gels, ie, they are highly hydrophilic substances that dissolve or disperse in water by increasing the viscosity of the system. These materials originally obtained from plants and seaweed have been gradually replaced by polymers obtained by fermentation processes. The species of nitrogen fixing bacteria are generally producing EPS. Among these, species belonging to the families Rhizobiaceae and Phyllobacteriaceae synthesize large amounts of extracellular polysaccharides. Studies cited in the literature showed that with the advent of molecular biology techniques and advances in genome sequencing have allowed a new approach for elucidating gene functions, by evaluating the phenotypes of mutations by obtaining from a specific gene. In this project we intend to obtain mutants in the gene exoZ wild Rhizobium etli strain LMG 17 827 involved in the synthesis of the repeating subunits of EPS. The mutants with the desired gene will be eliminated constructed by insertion of the transposon EZ:: TN " , and subsequently will be conducted comparative analysis of phenotypes of wild type and mutated for new potential biotechnological applications.
