Protease activity of podosomes in a cell line derived from human adenoid cystic carcinoma

Abstract

Podosomes are adhesion structures found in different cell types, such as fibroblasts, lymphocytes and malignant cells. They are composed by a core of actin surrounded by a ring of the actin-binding proteins talin, paxillin and vinculin. Since podosomes express matrix metalloproteinases (MMPs), they are probably related to cell invasion. Our Laboratory have been studying the biology of salivary gland tumors. Our results show that a cell line (CAC2) derived from adenoid cystic carcinoma express podosomes. We then decided to study the role played by podosomes on protease activity of CAC2 cells. The investigation will be carried out using in situ zymography and immunofluorescence. CAC2 cells will be grown on gelatin-FITC for 5, 10 and 15 minutes. Digestion regions will be shown as dark areas in a green fluorescent background. The same cells will be double-labeled to detect MMPs and vinculin (typical podosome protein). The colocalization of dark areas of digestion with vinculin and MMPs will characterize protease activity of CAC2 cells. (AU)
