Abstract

This project aims to study the molecular involvement of dusp12 gene in mouse embryonic fibroblasts (MEp) isolated and established from dusp12 gene targeted mouse. The absence of dusp12 gene in cell cycle control proliferation and migration of MEF lines will be investigated. DUSP12 protein expression will be checked as well as phosphorylation (activation) of MAPKs in parental MEFs ou isolated MEF lines over expressing dusp12. Responsiveness; of these MEF lines to FGF2 stimulation will also be investigated as to proliferative and/or antiproliferative FGF2 effects. In order to identify specific substrates or partners of DUSP12 protein to perform its cellular actions, a yeast-two-hybrid screen will be made and identified genes will be over expressed or knockdown by siRNA in MEF and other mouse lines.
