Evaluation of intraovarian application of mesenchymal stem cells and conditioned medium in bovines

Abstract

The aim of this study was verify whether intraovarian applications of mesenchymal stem cells and conditioned medium in bovines are safe techniques, not causing a pathological inflammatory process. Twelve non-lactating Jersey cows were used with an average age of 5.7 years and average BCS of 2.5 points, belonging to the Faculty of Social and Agricultural Sciences of Itapeva – FAIT in the municipality of Itapeva-SP. The experiment was paired: the same animals were used at three different moments and each stage constituted an experimental group. Control group: in the first stage all animals received an intraovarian application in both ovaries of 1 mL of control medium (PBS) per ovary. MSC group: in the second stage the same animals from the control group received in each ovary five million MSCs diluted in 1 mL of medium. CM group: subsequently in step three 1 mL of CM of MSC was applied to each ovary of the same 12 animals from the first and second stages. Analyzes pre- and post- intraovarian applications were performed at several moments, evaluating: hematological values, fibrinogen dosage, body temperature, serum AMH concentration and ovaries ultrasonography. The results showed there was no difference in body temperature (p= 0.4347) and in fibrinogen concentration between groups(p=0.0844); there was a difference between the control and the MSCs groups in the total number of leukocytes (p=0.0149), with the highest value observed one day after the application of MSCs (15188.4 ± 1632.5 leukocytes/mm³); there were no alteration in AMH concentrations between groups (P = 0.3081); the subjective analysis of the ultrasound examination showed that no animal developed significant changes in the patterns of echogenicity of the ovaries. In conclusion, the MSCs and the CM when intraovarian applied did not lead the animals to a local or systemic pathological inflammatory process. (AU)
