Microbiological investigation, growth condition, and metabolic potential for research of biactive natural products on endophytic fungi Cladosporium sp (AC-1) isolated of Annona cacans (Annonaceae)

Abstract

This study aimed to investigate microbiological parameters, growth conditions and the metabolic potential, aiming the search for bioactive natural products of endophytic fungus AC-1 isolated from Annona Cacans (Annonaceae). The isolation and identification of fungus AC-1 were performed using conventional microbiological analysis focusing on the macroscopic and microscopic morphology and ITS region of the ribosomal DNA, being suggestive results that the fungus endophyte AC-1 belongs to the genus Cladosporium sp., specifically C. cladosporioides. For the extraction of secondary metabolites were used sources nutricionales different and as extracting solvent is ethyl acetate (EtOAc). The production micromolecular metabolites of these extracts was investigated by GC-MS, HPLC-UV/DAD, ESI-MS/MS and UHPLC-MS and evaluated their potential as the antimicrobial activity (against bacteria and species Candida spp) and antioxidant (DPPH methods and ORAC-FL) for the expansion of the scale, fraccionamiento, isolation and identification of bioactive substances is selected from the extract through YM medium (extract with the best results in terms of activity and in mass yielded). For the isolation and identification as a result a pure substance was identified trans-cinnamic acid (substance 1) and two substances identified as a mixture, the Rel.cyclediketopiperazine (Pro-Ile) (substance 2) and Rel.cycle (Pro-Phe) (substance 3), whereas other purified fractions are still being analyzed by NMR. Also were identified m/z suggesting the possibility of the presence of asimilobina, anonaina, liriodenine, coumaric acid and cinnamic acid that have been reported in species of Annona spp. and cladosporol B, cladosporol D, cladosporina, recifeiolide and fatty acids that have been documented in fungi species of the Cladosporium. The extracts and fractions showed promising results in the antimicrobial and antioxidant assays and were identified substances with proven antimicrobial and antioxidant activities, including anonaine, asimilobine, trans-cinnamic acid and the diketopiperazines 2 and 3. The continuity of chemical and biological research is necessary aiming to confirm these results and evaluate the best biotechnological potential of this endophytic microorganism (AU)
