Influence of isotretinoin on orthodontic tooth movement in rats: microtomographic and histomorphometric analysis

Abstract

Isotretinoin, or 13-cis-retinoic acid, is an isomer drug of tretinoin, the oxidized form of Vitamin A, and is part of the most common retinoids of approximately 1500 synthesized. Orthodontic tooth movement (OTM) is the result of a cascade of inflammatory responses stimulated by a physical element that is the force generated by orthodontic appliance, involving the remodeling of the alveolar bone, alterations in the periodontal ligament and sometimes in the root cementum. Isotretinoin is used mainly among adolescents and young adults, and coincidentally it is the age group that also undergoes orthodontic treatment. In this study, we used an animal models (n= 55) to observe the effects of isotretinoin on OTM in rats. After euthanasia, the animals' jaws had been surgically extracted and subjected to histological examination and analysis by Micro-CT. Specific statistical tests were used to analyze whether there was a difference in tooth displacement and also in the microtomographic parameters of animals submitted concomitantly to isotretinoin and OTM. Our results showed that isotretinoin influenced OTM on day 14, with a dose of 7.5mg/kg/day. (AU)
