Evaluation of the transfer of cytokines to the blood of neonates by colostrum ingestion of Holstein females

Abstract

The birth of a calf is marked by the passage of a sterile environment to a contaminated environment, coupled with the fact of placentation (sindesmocorial), which does not allow the passage of antibodies during pregnancy and without an immune system developed functionally, the needs of the neonate ingestion of colostrum to give it immunity during the first days of life. Besides the colostrum immunoglobulin has leukocytes and cytokines that seem to participate in the establishment of neonatal immunity. Despite the potential importance of these components of colostrum , there are few studies evaluating the transfer of cytokines from colostrum and its role in neonatal immunity, reason why this project had the following objectives: to measure the concentrations of IL- 1β, IL- 6, TNF-α and IFN-y in the serum of newborn Holstein calves and evaluate the interference of bovine colostrum management and of his offering , or not , on the concentrations of these cytokines in the serum of these calves during the first 15 days after birth. We used calves born from normal delivery and equally divided into three experimental groups (n = 5): G1 - received 4 liters of fresh colostrum from their mothers; G2- received 4 liters of colostrum from a \"pool\" of frozen colostrum and G3- were fed only milk. For the evaluation of these cytokines in the serum of calves, blood collections were made during the first fifteen days of postnatal life (p.n.): T0 (before the first intake of colostrum or milk), T1 (1st 2 days p.n.), T2 (3 to 4 days p.n.), T3 (5 to 6 days p.n.) and T4 (10 to 15 days p.n.). Also measured is the cytokine TNF-α , IL-1β and IL-6 in the supernatant of colostrum fed to calves in group G1, and cytokines TNF-α , IL- 1β, IFN-y and IL -6 in supernatant \" pool\" of colostrum fed to calves in G2. Calves from G1 group had detectable values for these cytokines, reaching peaks at T1 (2 to 3 days p.n.) or T2 (3 to 4 days p.n.) while calves in G2 showed detectable values only for the period T3 ( 5 to 6 days p.n. ) for the cytokine IL-6, whereas the calves of group G3 did not show any detectable for cytokine concentrations measured with the exception of IL- 1beta 4 time ( 10 to 15 days p.n.). Based on findings of this study it was possible to: (a) verify the transfer of IL- 6, IL- 1β, TNF-&#945 and IFN-y in the serum of calves from G1 via ingestion of fresh colostrum. (b) verify the transfer of cytokine IL -6 into the circulation of calves in G2 via ingestion of \"pool\" of colostrum. (c) verify that deprived newborn calves colostrum (G3) showed no detectable concentrations of IL - 6 , IL- 1β , TNF-α and IFN-y in the first fifteen days of life , demonstrating that detectable concentrations of cytokines in question come from the colostrum. (d) verify that the group of calves that received colostrum from their mothers (G1) had higher cytokine concentrations than the group that received \"pool\" of colostrum (G2), demonstrating that leukocytes present in fresh colostrum are able to express cytokines in peripheral blood of newborns up to 15 days after birth calves.
