Spin state characterization and aggregation of microperoxidase-8, -9 as a function of pH and pulsed EPR of cytocrome c (S=½)

Abstract

In this work we have used spectroscopic techniques such as UV-vis, circular dichroism, and electronic paramagnetic resonance to characterize the microperoxidases-8 and -9 and also N-AcMP-9. Microperoxidases are products of the proteolytic digestion of cytochrome c, containing an average of ten residues of aminoacids. Lacking the sixth ligand, the heminic iron is ín the high spin state. Our results show that the alkaline transition (pKa = 8,53) produces hemepeptideos aggregates in the low spin state, in which the water molecule is displaced by the Nterminal group of a nearby MP-8 cysteine. For high values of pH there is a deprotonation of the imidazole ring of the histidine that occupies the heminic iron\'s fifth coordination. In that pH MP-8 is found in equilibrium of aggregated species of high and low spin. An aggregated species if formed by the pentacoordinated MP-8 with the histidinate in the sixth coordination. The other aggregated species is composed by the coordination of the alpha-amino groups of cysteine and hydroxide ion, occupying the sixth and fifth coordination of heminic iron, respectively. As for NAcMp-9, the low spin state is produced by deprotonation (pKa=9.6) of the water located at the iron\'s sixth coordination. These results agree with others found for NAcMp-8. In MP-9 the low spin state is obtained displacing the water by the terminal alpha-amino group of Lysl3 in a nearby molecule. Above pH 8.5 the epsilon-amino group of Lysl3 coordinates intra-chain. The monomers exhibit OH- or histidinate in the fifth coordination. The intra-chain coordination of MP-9 was confirmed by molecular dynamics simulation. It is important to notice that all results from CD and EPR were simulated using the programs CCA and QPOW, respectively. We also made pulsed EPR measures of cytochrome c in fosfato buffer pH 7.4 in SDS and in cardiolipin, in order to investigate the alternative low spin state. We made experiments with echo field sweep, ESEEM and spin-lattice relaxation time TI for a complete characterization of the above mentioned mixture (AU)
