(EN) The present invention relates to a vector, called pMF_002 vector, and to the method of separating and collecting S. cerevisiae haploids using this vector in order to express fluorescent proteins (markers) in haploid (MATa and MATα) cells and thus distinguish them from each other and from tetrads and diploids, using a flow cytometer. For that purpose, this vector comprises the reporter genes EGFP (Enhanced Green Fluorescent Protein) and CyOFP1 (Cyan Orange Fluorescent Protein), the promoters STE2 (STErile2) and STE3 (STErile3), the terminators CYC1 (Cytochrome C1) and IDP1 (Isocitrate Dehydrogenase 1), and the backbone vector (pGS_004).
