Controlling Alicyclobacillus acidoterrestris spores contamination on orange surfaces using different photosensitizers and light sources

Abstract

This study aimed to assess the use of antimicrobial photodynamic treatments (APDT) to control Alicyclobacillus acidoterrestris spores on orange surfaces. The minimal inhibitory concentration (MIC) assays revealed that 125 mu M of new methylene blue N (NMBN) + red-LED for 180 min, 160 mu M of curcumin (CURC) + blue-LED for 180 min and 160 mu M of 8-methoxypsoralen (8-MOP) + solar radiation for 120 min were inhibitory for A. acidoterrestris. The APDT applied to oranges using 8-MOP and solar radiation resulted in 1.6 log spores/orange reductions of A. acidoterrestris. The APDT with NMBN and CURC using a device ("photodynamic box") resulted in 0.7 and 0.8 log spores/orange of A. acidoterrestris, respectively. The results show the potential of APDT technology for in-field and pre-processing decontamination of A. acidoterrestris spores on oranges. Implementing APDT on-farm and at the initial steps of juice processing could radically change the control of A. acidoterrestris in the fruit juice industry. (AU)
