Mechanistic study of perception and response to visible light in the entomopathogenic fungus Metarhizium acridum

Abstract

Visible light is an environmental factor that regulates metabolism, growth, reproduction and even pathogenicity and virulence in different species of fungi. Exposure to light can also pre-adapt fungal structures, such as conidium and mycelia, to future conditions of stress. In the entomopathogenic fungus Metarhizium acridum, exposure to visible light increases the tolerance to UV-B radiation (280-320 nm). As Metarhizium is used as mycoinsecticide the induction in tolerance to UV is highly desirable, since one of the factors that compromise its effectiveness is the inactivation of the inoculum by solar radiation. Despite the practical importance that the induction of UV tolerance has for the use of Metarhizium spp as mycoinsecticides, the molecular mechanisms that result in this effect have not been determined. The overall objective of the present project is to elucidate the molecular basis of the induction of UV tolerance by visible light in Metarhizium. The identification of the photoreceptors involved in the perception of light will be made by obtaining fungal strains delected to the genes for the major known photoreceptors (white collar-1, Cry-DASH, cryptochrome-2 and photolyase). Additionally, transcriptomic by deep sequencing of RNA (RNA-seq) and proteomic by LC-MS / MS approaches will be used in an integrated and complementary way, on the same samples, to identify genes and proteins involved in the response to visible light. It is expected that the results will provide information on the molecular mechanisms involved in the perception and response to light and also contribute to the production of M. acridum strains more tolerant to environmental stress, and therefore more suited to be used as micoinseticidas.
