Ionic Liquids as Green Extractors for Achievement of Carotenoids: Study of Bioavailability and Antioxidant Activit

The application of ionic liquids (IL) in extraction processes is a viable alternative, which follows the principles of green chemistry, minimizing contamination and optimizing the achievement of these natural compounds. Thus, the aim of this study will be establish a new method of extraction using IL for achievement of carotenoids from orange peel and pupunha pulp (Bactrys gasipaes), which can be used as natural pigments and additives in the food industry. Four different IL will be tested as extractors, the 1-n-butyl-3-methylimidazolium tetrafluoroborate and the 1-n-hexyl-3-methylimidazolium chloride from trademark; and the 1-n-butyl-3-methylimidazolium chloride and the 1-n-butyl-3-methylimidazolium hexafluorophosphate synthesized in laboratory. The synthesis assay of the IL will be assisted by ultrasound, obtained without the use of harmful organic solvents, and the characterization of the IL will be performed by mass spectrometry (MS). Preliminary tests will be conducted for select the most effective IL for extraction of carotenoids. The efficiency parameter assessed will be the yield in extraction of carotenoids (quantification by external calibration curve in HPLC-PDA). The two best IL will be selected to optimize the extraction using the response surface technique, through a fractional factorial design, followed by a full factorial design. The qualitative and quantitative determination of carotenoids extracts will be performed by HPLC-PAD-MS/MS. The stability of carotenoids extracts obtained by conventional extraction and by IL will be assessed in relation to the presence and absence of light and at different storage temperatures. For carotenoids extracts, stability will be further evaluated in two distinct systems: oily and watery. The bioavailability in cell model system (Caco2) and antioxidant activity will be compared with carotenoid extracts obtained by conventional methods employing acetone as solvent extractor. With these results we intend to advance in the study of the properties of ionic liquids in obtaining bioactive ingredients, enabling the development of processes with less waste, and with the possibility of patent filing.
