Analytical Approaches for Discovering Unknown Cytotoxic Compounds from Plants

Abstract

Natural products (NPs) have been part of drug discovery process, including cancer therapy. However, a constant hurdle in cancer area is drug resistance. Alternatively, treatment strategies can be combined to control or inhibit the proliferation. In executing such strategies, Science observes the return of NPs discoveries, after decades of focus on synthetic chemistry. The advances regarding analytical tools involving hyphenated techniques have boosted the find of new NPs from nature, what was provided by Metabolomics tools that allow rapid access to structural features. Publications suggest that the most promising workflows for the chemical profiling of NPs should involve a strategy able to distinguish known and unknown compounds, e.g., the dereplication approach. Based on this, the main goal of our proposal is the use of modern analytical techniques for finding unknown and known compounds from plants guided by cytotoxic assays against four human leukemia cells and also solid tumors. A collection of 90 crudes extracts, prepared in our lab was submitted to antiproliferative screening during 24 hours at 100 mg/mL. Results indicated that 30 samples have presented activity at least against one leukemic cell, and also were viable when assayed on non-leukemic cells. Active samples will be dereplicated by chromatographic and spectroscopic methods for the preliminary recognition of the bioactive compounds (AU)
