Abstract
The purification of active components from venoms with high purity and in relatively high amounts is a limiting factor for the success of this project, since these active molecules are the object of study of all the proposed tests, aiming at obtaining compounds with biotechnological potential. Prominence chromatography system is a modern equipment and meets all requirements for the isolation of pure toxins, because it works with high pressure, can be used with columns of different characteristics (ion exchange, reverse phase, etc.), both preparative or analytical. In addition, it has ample options for the type of buffer to be used, which can alter composition, ionic strength and even pH during the purification process. Absorbance data at different wavelengths and number of tubes are automatically obtained during the purification process. The Prominence Chromatography System will be used for purification processes and to meet the demand for fractionation of peptides obtained by proteolysis of proteins submitted to PPSQ-53A sequencing and for synthesis. This is a process that requires a long time because, depending on the size of the protein, a large number of peptides can be obtained, which would require further isolation and sequencing. Its acquisition is therefore fundamental for the processes of obtaining and characterizing proteins of biological interest to be viable and efficient. (AU)
