Antimicrobial resistance factors: characterization of AmpC, KPC and MCR-1 in resistant bacteria in the productive chain of broiler chicken

Abstract

Brazil is the major exporter and the third largest producer in the world of chicken meat, and it makes the requirements are strict, especially about the quality of the exported product. Any problem in this sector can be a risk for Brazilian agribusiness. Besides the importance of the poultry industry in the country, it is known that the use of antimicrobials as growing promoter in this sector, can be the major responsible about the increasing of infections caused by resistent Enterobacteriaceae to a wide variety of antimicrobials. And the biggest problem is because the emergence of new therapeutic resources does not follow the evolution of resistance bacteria mechanisms, become the infections caused by these strains a serious public health problem. Based on this, the present work aims to detect and characterize molecularly the presence of the main and most current resistance factors, namely: resistance to betalactams (AmpC) carpabenems (KPC) and colistin (MCR-1) in enterobacteria isolated in the whole poultry production chain, including animals, food and consumers. After phenotypic screening for antimicrobials, the genotypic detection will be performed by PCR and sequencing. Furthermore, characterization through the phylogenetic grouping as well as the clonal profile of the isolates and genes investigated by PFGE and MLST will be defined. The results of this study will be crucial to understanding the profiles of the most frequent resistance genes in broiler production chain in São Paulo state, and also will show the correlation of the presence of resistance genes in animals and humans.
