Abstract

Recently, a high-fat diet has been associated with hypothalamic low-grade inflammation, and dysregulation of energy and metabolic control that can lead to the development of obesity. In this process, the connection between gut and brain seems to be involved in the alterations that lead to the development of chronic diseases. Considering the evidence that an inadequate maternal diet, and more recently a paternal diet, can program the offspring to develop chronic diseases into adulthood, it will be assess the effects of maternal and paternal high fat/high sugar on the programming of the hypothalamus and colon from the 90-day old offspring. Therefore, it will assess in the 90-day old offspring from parents who had high fat/high sugar or control diets, the following parameters. Serum fatty acids profile; hypothalamic and colonic relative weight; leptin and insulin content in the hypothalamus; cytokines TNF-±, IL-6, IL10 and LPS content in the hypothalamus and colon; TLR4 content and proteins involved in the TLR4 pathway MYD88, NFkB fosforilado (p50 e p65) content in the hypothalamus and colon; ZO-1 content in the hypothalamus and colon; quantification of Lactobacillus and Bifidobacterium DNA in the stools.
