Effects of oral administration of oleic and linoleic acids on experimental Psoriasis: focus on skin dendritic cell functions

Abstract

The skin is an immunologically organ where unregulated immune reactions can cause chronic inflammatory diseases such as psoriasis that affects thousands of people worldwide; It has a relapsing-remitting character and may be accompanied by diseases such as obesity, diabetes, dyslipidemia and depression. Activation of dendritic cells plays a central role in the induction and maintenance of the disease. Oral administration of fatty acids has been a successful strategy for treating inflammatory diseases. However, few studies have investigated the effects of oral administration of oleic and linoleic fatty acids on dendritic cell functions and their consequences on psoriasis. Thus, the objective of the present study is to evaluate the effects of oleic and linoleic fatty acid intake on psoriasis, focusing on the functions of skin dendritic cells. In vivo, mice will receive oleic or linoleic acid for 6 days and then psoriasis will be induced from the 6th with the daily imiquimod (IMQ) topical application until the 13th day. The animals will be euthanized for collection 9, 11 and 13 days after the beginning of the protocol. The macroscopic and microscopic effects of supplementation will be evaluated, as well as the cells (by flow cytometry) in the skin and the cytokines and chemokines (by ELISA) in the serum and skin of the mice. Gas chromatography coupled with mass spectrometry will evaluate the incorporation of fatty acids in serum and skin. The effects of supplementation on cellular immunophenotyping of skin, spleen and lymph nodes will also be investigated. After determining the main altered cell populations in each of the process phases, these cells will be isolated by FACS Sorting for RNASeq Single Cell gene expression analysis. We will use epidermal and dermal dendritic cell depletion protocol to investigate whether the effects of fatty acid supplementation during psoriasis are dependent on dendritic cells. In vitro, we will analyze the effects of oleic and linoleic acid on cytokine production, activation, maturation and migration of dendritic cells isolated from bone marrow of C57BL/6 mice. (AU)
