Potentiation of effects induced by red laser induced by herbal medicines

Abstract

Literature papers have shown that the red LASER (660 nm) is capable of inducing vasodilation and accelerating the wound healing process by a NO-dependent mechanism. Furthermore, it is known that ATP promotes vasodilation via receptors of the purinergic system, with the participation of NO, and that LASER stimulates its production via Cytochrome C oxidase. Therefore, increasing mitochondrial biogenesis from the synergistic action of Angelica keiskei and Panax ginseng extracts becomes an interesting strategy to potentiate the effect of LASER. Therefore, the objective of the project is to investigate the use of herbal medicines as a strategy to potentiate the biological effects induced by the red LASER. The proposal is to test this strategy in two experimental models, in which the red LASER has known effects, such as the hypotensive effect and the acceleration of wound healing. Also, it will be verified if this potentiation occurs in isolated vessels, removed from patients after bariatric surgery, from the previous treatment of isolated vessels with herbal medicines. For this project, in vitro studies will be carried out: such as the analysis of cytotoxicity and cell proliferation of endothelial cells and fibroblasts (via MTT) from the association of herbal medicines with photobiomodulation, analysis of transcription factors related to mitochondrial biogenesis and inflammation, by Western Blotting , as well as the collection of blood from the animals for analysis of serum ATP levels; the in vivo studies, in 2K-1C and normotensive rats with wounds, will be divided into experimental groups where each group will receive a different treatment (LASER, Phytotherapics and LASER + Phytotherapics association) and the blood pressure levels of these animals will be evaluated; wound closure; there will also be a study on isolated aortic rings, in which protocols will be carried out to study the endothelial function of the different groups, the mechanism of action of the potentiating effect and the potentiation action in vessels removed from patients after bariatric surgery. Our hypothesis is that the association of herbal medicines A. keiskei and P. ginseng potentiates the biological effects induced by the red LASER, which may contribute to clinical practice, allowing greater effectiveness of treatments, including the acceleration of wound closure that can contribute to relieve the public health system.
