Effects of aerobic exercise training on perivascular adipose tissue in obese female mice: evaluation of vascular function and biomarkers of inflammation, oxidative stress and senescence

Abstract

The perivascular fat tissue (PVAT) plays an important role in controlling vascular tonus and aerobic exercise directly interferes with the secretory pattern of this tissue. Pro-inflammatory factors secreted by PVAT associated with oxidative stress contribute to the development of vascular complications in obesity. The mechanisms responsible for the dysfunction of PVAT in obesity are not yet fully elucidated, being most of the studies performed in males. Thus, this study will aim to evaluate the effects of aerobic physical training on PVAT in obese female mice. Female mice C57BL6/JUnib will be fed a standard or high-fat-diet and submitted to moderate intensity aerobic training protocol. Concentration-response curves to acetylcholine, sodium nitroprusside, thromboxane analogue A2 and phenylephrine will be performed in thoracic aorta rings in the presence and absence of PVAT. In aorta the expression of proteins will be evaluated: eNOS, peNOSser1177, AdipoR1, APPL1, AMPK, ObR, p53 e p21; and in PVAT eNOS, iNOS, APPL1, UCP-1, p56 and p21. The production of NO and generation of reactive oxygen species (ROS) will be determined in arteries and PVAT. The senescence cells will be quantified with beta-galactosidase staining. Serum levels of glucose, leptin, TNF-alpha, adiponectin, TBARS, SOD and catalase activity and 17beta-estradiol will be evaluated. In the PVAT it will be realized histological analysis, of the leucocyte infiltrate and the profile of polarization of the macrophages. (AU)
