The perivascular adipose tissue (PVAT) plays an important endocrine and paracrine role and aerobic exercise directly interferes with the secretory pattern of this tissue. Pro-inflammatory factors secreted by PVAT associated with oxidative stress contribute to the development of cardiovascular complications in obesity. The mechanisms responsible for PVAT dysfunction in obesity are still not fully elucidated, with most studies performed in males. Thus, the present study will aim to evaluate the effects of aerobic exercise training on inflammatory, oxidative stress and senescence biomarkers of PVAT in obese female mice. C57BL6/JUnib female mice will be fed a standard or high-fat diet for 16 weeks, and submitted to moderate aerobic exercise training on a treadmill for 8 weeks. In the PVAT of the thoracic aorta (tPVAT), histological analysis will be performed for morphological characterization of the tissue and the leukocyte infiltrate and the macrophage polarization profile will be evaluated. The expression of proteins eNOS, iNOS, APPL1, UCP-1, p53 and p21 and the generation of reactive oxygen species (ROS) will be evaluated in tPVAT. Quantification of senescent cells will be performed by ²-galactosidase assay. The serum levels of glucose and 17²-estradiol will be measured, as well as, TNF-±, adiponectin and TBARS in tPVAT.
