Effects of different experimental titanium alloys on proliferation and osteogenic potential of humane osteoblast in vitro

Abstract

The regular grant approved by FAPESP 2022/007162-5 aims to evaluate the chemical and tribochemical properties of experimental titanium alloys containing tantalum and zirconium metals, with potential applications as dental implants However, for the real applicability of these new materials in clinical practice, biocompatibility studies must be carried out to verify the biological responses of the material. Thus, this project will clarify the best surface of experimental titanium alloys in relation to cell culture, contributing to a better selection of dental material to be used in in vivo that can be used in research in future collaborations with the German research group.The Department of Cranio-Maxillo-Facial Surgery at the University Hospital of Münster has been internationally recognized for its scientific research in the field of vascular biology of oral structures (VABOS) (Happe et al., The Biologic Effect of Particulate Titanium Contaminants of Dental Implants on Human Osteoblasts and Gingival Fibroblasts. Int J Oral Maxillofac Implants. 2019; 34(3):673-680; Eggers et al., Effect of Cold Atmospheric Plasma (CAP) on Osteogenic Differentiation Potential of Human Osteoblasts. Int J Mol Sci. 2022 24;23(5):2503.) The VABOS Research Group is a renowned interdisciplinary team that includes physicians, molecular biologists, biologists, and physicists. Research is conducted in the field of bone regeneration, dental implants, cellular and molecular tissue regeneration, and angiogenesis.In this way, the exchange with the University of Münster in Germany will allow monitoring the cell response on the osseointegration process in contact with the new formulated alloys based on titanium, which will promote a considerable scientific addition to the regular project previously approved by FAPESP, allowing the beginning of collaborations between the two institutions. Likewise, it will allow the University of Münster access to new titanium alloys with a view to possible clinical applicability. Thus, with the approval of this proposal, Prof. Dr. Kleinheinz´s and Prof. Dr. Galo´s group will investigate, for the first time, the biological response of innovative titanium alloys prepared after tribochemical degradation with the cell culture assays for oral rehabilitation through dental implants. As the focus of this research is to restore the masticatory function in order to delay the aging of the stomatognathic system, dental implant alloy play an important role on the treatment. Our purpose is to associate, by means of a multidisciplinary approach, the ongoing investigation of physic mechanical properties to the biological compatibility of experimental dental alloys with human tissues. For this purpose, the effect of titanium alloy on osteoblastic proliferation and differentiation will be assessed. Cytotoxic properties of different material surface treatment, such as the effects on the osteogenic potential will be analyzed by quantifying the alkaline phosphatase rate and mineralization potential.Therefore, this FAPESP/SPRINT project is a valuable opportunity to begin the cooperation between the Department of Dental Materials/Biomaterials of the University of São Paulo and the Department of Cranio-Maxillofacial Surgery of the University of Münster. The current regular support from FAPESP, in association with the SPRINT-Program, will serve as a platform for sharing knowledge, training human resources through the exchange of students and promotion of research and education activities, also contributing to internationalization.
