Abstract

Intracanal medications must have adequate physicochemical properties and antimicrobial activity. Calcium silicate bioceramic materials are developed from tricalcium silicate (STC, IFUSP, São Carlos - Brazil). Biosilicate (BS, LaMaV/UFSCar, Brazil) is a glass-ceramic with the ability to promote osteogenesis and antimicrobial effects. Essential oils can improve the antibacterial effect for medications. This study is divided into 4 subprojects: Subproject 1: It aims to evaluate pH, solubility and antimicrobial activity by comparing different intracanal medications. (A) Bio-C Temp bioceramic medication (BCT, Angelus, Brazil); experimental tricalcium silicate paste (STE): STC, zirconia oxide (ZrO) as radiopacifier and polyethylene glycol (PL) vehicle; experimental calcium hydroxide paste (HCE): (HC + ZrO + PL); STE associated with 50 and 75% HCE. The effect of intracanal medications on the bond strength of bioceramic and resin cements will also be evaluated. (B) experimental BS paste (BSE): BS + ZrO + PL; HCE experimental calcium hydroxide paste: HC + ZrO + PL; BSE associated 50 and 75% HCE paste. Bond strength after medication and use of endodontic cements will also be evaluated. (C) experimental tricalcium silicate medication- STE; HCE and its associations with Thyme oil (Theo): STE + Theo 5%; HCE + Theo 5%. (D) experimental tricalcium silicate medication - STEXP; HCE and its associations with carvacrol (CAR): STE + CAR 5%; HCE + CAR 5%. Subproject 2: Aims to develop and evaluate physicochemical properties, bioactive potential and antimicrobial activity of an experimental bioceramic endodontic sealer based on STC (IFUSP, São Carlos, Brazil): (CBEXP) in comparison to Bio-C Sealer (Angelus, Brazil) and AH Plus (Dentsply - Switzerland). Subproject 3: Aims to evaluate the treatment of gutta percha (GP) by biosilicate using surface coating method (dip-coating) and incorporation into GP. Subproject 4: Aims to evaluate the biocompatibility and bioactivity of the main intracanal medications from subprojects 1 to 4, the endodontic sealer developed in subproject 1 and the GP treated by biosilicate (subproject 3) by using analysis in rat subcutaneous tissue. Volumetric change will be assessed using microcomputed tomography (micro-CT) (Subproject 2). Bioactive potential will be evaluated using scanning electron microscopy (Subproject 2). Antimicrobial activity will be evaluated using the modified direct contact test, confocal laser scanning microscopy (Subproject 1-2) and decontamination of contaminated roots in vitro by colony forming unit counting and action on LPS, by the LAL test (Subproject 1). The data will be subjected to appropriate statistical tests (±=0.05). (AU)
