Interactions of ascorbate bioflavonoids and redox biofactors in cytochrome c - induced peroxidative stress and apoptosis.

Abstract

We propose investigations to extend preliminary observations that the bioflavonoid epicatechin rescued protection by ascorbate against peroxide-induced bleaching of cytochrome c that was prevented by the presence of liposomes of mitochondrial membrane-like composition. To the extent that the ascorbate-mediating effect of epicatechin is specific to one or more bioflavonoids and holds true in mitochondria and in cells, it suggests a vital cooperative activity of a bioflavonoid not previously seen in studies of vitamin P activity. Objectives of the investigations are to determine the prevalence of the effect among other bioflavonoids, and the influences of this interaction on cytochrome c-catalyzed peroxidations of liposomes and mitochondrial membranes. In addition we propose experiments to investigate the influences of peroxides, ascorbate, bioflavonoids, and other redox factors on the newly found pathway of apoptosis triggered by cytochrome c release from mitochondrial membranes.
