Abstract

The group IB corresponds to pregnant women bearers of mild hiperglycaemia, in other words, pregnant that present positive screening, but negative diagnosis for diabetes normal gestacional(OGTT100g), and answer altered in the glycaemic profile (GP). This group was, accidentally, identified in 1983, when I project prospective it was developed for standardization of PG comparing him/it with OGTT 100g in the diagnosis of the diabetes in the gestation (GDM). Those pregnant ones present resistance to the insulin, intolerance to the glucose, larger susceptibilidade of developing diabetes type 2 some years after delivery, and 53,8% of their newborn they are big for age gestacional and/or macrosomics, similar to the women with GDM. The objective of that work is to evaluate the picture of light hiperglycaemia in the gestation, to identify molecular biomarkers and possible related genetic alterations the that abnormal glycaemic. They will be appraised women assisted in School of Medicine of Botucatu - São Paulo State University, UNESP between 37 and 42 weeks of gestation. The patients will be divided in 4 groups: 1) women with normal OGTT and GP (control group), 2) women with normal OGTT and abnormal PG (mild hiperglycaemia group), 3) women with abnormal OGTT and normal PG (gestational diabetes group), 4) women with abnormal OGTT and PG (overt gestational diabetes group). Samples of blood, placenta and umbilical cord blood will be collected by experienced professional and stored for subsequent extraction of DNA and RNA. They will be appraised the one of polimorfisms of the genes: PPAR-gamma, INS, ADIPOQ, besides the gene expression: PPAR-gamma (blood), TNF -, adiponectin and placental lactogen (placenta). it will be made the maternal dosage of insulin, adiponectina, TNF -, placental lactogen and free fatty acid concentrations to correlate with the polimorfisms and gene expressions. The significance level for all test will be considered <0,05. (AU)
