Time-course of oxidative stress induced by doxorubicin in rats: damage mechanism

Abstract

DOX is excellent drug in treating human cancer; however, the induction of cardiotoxicity has limited its use. Oxidative stress (OS) has been a common denominator in the DOX-induced cardiotoxicity mechanism. However, our laboratory has showed that chronic experimental design (which mimics the clinical situation) presents an antioxidant feature of doxorubicin rather than acting as a prooxidant. The aim of this study is monitor the OS during the DOX therapy. Wistar-rats will be randomly assigned to either a Control or DOX groups. The Control group will receive intraperitoneally (IP) saline, once a week/4weeks. DOX group will receive DOX (IP) 4 mg/kg instead saline. The animals will be killed 24h before and after the 1st, 2nd, 3rd and 4th IP injections (acute damage) and also one and four weeks after the final DOX therapy (chronic damage). The myocardium EO will be monitored by: morphological exam, total antioxidant performance, malondialdehyde and DNA oxidative damage. (AU)
