Evaluation of the effect of Gamma-Oryzanol treatment on oxidative damage lipid and protein carbonylation in acute cardiotoxicity induced by doxorubicin

Cancer is a major public health problem worldwide. Among the treatments used, Doxorubicin (DXO) stands out, a drug used to treat various types of tumors. Despite its effectiveness, DXO has cardiotoxicity as an adverse effect. It is classically accepted that oxidative stress plays a role in the damage mechanism of secondary toxicity of DXO. In order to mitigate and/or prevent cardiotoxicity, several therapeutic strategies have been studied, among them, the use of bioactive compounds. Gamma-Oryzanol is known to have antioxidant activities. In this sense, the present study aims to evaluate the effect of Gamma-Oryzanol treatment on oxidative lipid damage and protein carbonylation on acute cardiotoxicity induced by doxorubicin in rats. Wistar, males (n = 32) will be initially randomized into two groups: Control group (C, n = 16), and Gamma group (G, n = 16), who will receive Gamma-oryzanol daily, diluted in corn oil, via gavage, in doses defined in a pilot project. On the 7th day, the animals will again be divided into four groups: Control (C, n = 8), DXO (D, n = 8), Gamma (G, n = 8), and Gamma + Dxo (GD, n = 8) all animals in the DXO groups will receive doxorubicin, intraperitoneal (IP) 4 mg/kg BW. 24 hours after the administration of doxorucicin the animals will be euthanized. For euthanasia, the animals will be anesthetized (via IP) with 200 mg/kg ketamine hydrochloride + 30 mg/kg xylazine and euthanized to obtain blood and heart for the analysis.(AU)
