Abstract

Mastitis is the most prevalent infectious disease among dairy cattle, and Staphylococcus aureus (S. aureus) is one of the most important causes of bovine mastitis. Molecular characterization of S. aureus isolates associated with bovine mastitis can be helpful in the development of more effective control practices of this disease. Pulsed-field gel electrophoresis (PFGE) has been considered one of the most reliable typing procedures, relevant in the development of mastitis control strategies directed to reduce the levels of herd infection. Besides, methicillin-resistant S. aureus strains makes these microorganisms one of the most important resistance mechanisms. Because of the importance to investigate the strains of S. aureus and due to the severe infectious and intoxications in man and animals that may produce, it will be collected milk samples of all the dairy herd in lactation, from ten farms located in several regions of Sao Paulo State. It will be realized microbiological analyses and biochemical identification from all isolated staphylococci from the animal teats and bulk tank, which will permit the classification in coagulase-positive and negative; sensibility profile of all the microorganisms isolated from the animal teats; identify genes responsible for the production of enterotoxins and toxic schok syndrome toxin 1 (TSST-1) in S. aureus and coagulase-negative staphylococci (CNS) isolated from the milk; detection of toxins A, B, C and D by reverse passive latex agglutination (RPLA) method and applicate the Pulsed-Field Gel Electrophoresis (PFGE) technique for genetic analysis of S. aureus isolates from teats and bulk tank.
