Structural proteome and characterization of ORF-31 product of the nucleopolyhedrovirus Anticarsia gemmatalis (AgMNPV-2D) in different cell lines

Abstract

Since the 80s the nucleopolyhedrovirus Anticarsia gemmatalis (AgMNPV) has been used in many Latin American countries and in the United States as a biologic control to the velvetbean caterpillar (A. gemmatalis). In Brazil there are at least two millions hectares of soybean plantation that uses viral control with great ecological and economical benefits. The AgMNPV also plays an important role as a clone vector and eukaryotic gene expression. Baculovirus are enveloped and belong to the baculoviridae family. They have a circular double-stranded DNA geome surrounded by a proteic capsid. At the end of the secondary infection, the virions become occluded inside a paracristalline matrix. The AgMNPV gene expression is temporally regulated. The expressed genes from distinct temporal classes regulate the expression of the following phase. Due to its great commercial and biotechnological importance, the baculoviruses have been thoroughly studied. Recently, our laboratory finished the AgMNPV-2D complete genome sequencing, finding 152 putative open reading frames (ORFs) including 3 unique ones, including ORF 31 that is trancribeed at the time of viral DNA replication an encodes for a poly-ADP-ribose-polymerase (PARP). As a next step after sequencing the AgMNPV genome, this project has as main goals: (i) determinate and characterize part of AgMNPV-2D structural protein sequence; (ii) identify the expressed proteins from the two viral phenotypes (ODV and BV), in distinct lepidopteran line cells; (iii) map in silico the ORFs encoding structural protein from both viral phenotypes; (iv) functional charaterization of ORF-31 PARP. This study will allow a better understanding of the AgMNPV morphogenesis and associated factors to viral multiplication. (AU)
