Abstract
JC virus (JCV) is the etiologic agent of progressive multifocal leukoencephalopathy (PML), a disease of the central nervous system of immunocompromised patients manifested by focal lesions without mass effect. The aim of the present study is to compare the prevalence of different genotypes of JCV in cerebrospinal fluid (CSF) and peripheral blood samples from patients with AIDS and PML and in blood samples from patients with AIDS without PML, matched with PML cases by CD4+ cells number. Samples will be obtained from AIDS patients admitted to three medical institutions (Instituto de Infectologia Emílio Ribas, Hospital das Clínicas da FMUSP e UNICAMP). Additionally, amplification and sequencing of the regulatory region of the virus strains obtained from the two groups of patients will be carried out. Detection of JC virus will be performed by two different PCR techniques (conventional and real-time PCR). In JCV positive samples the viral load will be determined by real-time PCR and molecular genotyping will be carried out by amplification and sequencing of the VP1 region of the virus genome. (AU)
