B3 adrenergic polymorphism and its correlation with overactive bladder syndrome and detrusor overactivity

Abstract

Overactive bladder is the most common bladder problem in late life.The underlying pathophysiology of the disease remain arguable. The most of the cases has an idiopathic nature. Because activation of the B3-AR causes a relaxation of detrusor muscle during bladder filling, we hypothesized that mutation of the B3-AR gene may also be involved in idiopathic overactive bladder, leading to an insufficient relaxation of bladder during urine storage, decreasing bladder capacity, and favoring unstable contraction. A miss-sense mutation in codon 64 of the B3-AR gene, with a replacement of tryptophan (Trp) to arginine (Arg) occurs with an important frequency in some populations. So, we proposed a case-control study to evaluate this polymorphism prevalence in overactive bladder syndrome cases (with or without urodynamic overactive bladder) compared with controls. We will study 300 women, 100 cases and 200 controls.Blood samples (5 ml) will be taken. Genomic DNA will be extracted from whole-blood leukocytes using a commercially available kit (GFX). Determination of the polymorphism will de done by polymerase chain reaction. All patients will be examinated and, only the case women will have urodynamic evaluation. (AU)
