Abstract

Metabolic flux analysis and other tools of metabolic and biochemical engineering will be utilized for the improvement of the poly-3-hydroxybutyrate-co-3-hydroxyalkanoate of medium-chain-Iength [P(3HB-co-HAmcl)] production. Flux analysis will be performed using a free access software and experimental data generated in bioreactor. The study of essential nutrients limitation to microbial growth, to induce accumulation of the target product, will be held in a continuous mode operation, employing the technique of multiple nutrients limitations. Metabolic models will be proposed, simulated and validated utilizing the experimental data from bioreactor cultivations. The analysis of the metabolic models will provide information for genetic modifications and process strategies, in order to obtain higher yields and productivities. It is expected that, by the end of this project, the metabolic engineering approach emphasizing the integration of different methodologies and procedures shall be established. As a result, a Metabolic Engineering Group will be created, in such a way that, processes involving others bioproducts could be approached in a similar manner. (AU)
