Abstract

Epoxide hydrolases are a group of hydrolitic enzymes free of co-factors. Such enzymes are widely spread in several organisms and act converting epoxides into the corresponding diols. Once this reaction could happen in a stereoselective fashion, epoxide hydrolases represent attractive asymmetric biocatalysts. Herein we propose the cloning and expression of Rhodotorula glutinis and Pichia stipitis epoxide hydrolases in Pichia pastoris, an efficient system for the expression of eukaryotic genes. After expression and chemical characterization of the enzyme catalytic activity we intend to exploit directed evolution approach combining appropriate methods for random gene mutagenesis/expression/screening or selection looking for enantioselective enzymes.
