Abstract

Application of enzymatic processes to the synthesis of homochiral intermediates possessing one or more stereogenie elements will be the main focus of this project. Monitoring microorganisms available in brazilian culture collections will allowthe detection of novel biocatalytic activities mainly from enzymes belonging to the monooxigenase, oxido-reductase, hidrolase and haloperoxidase type. The microorganisms (yeasts, fungi or bacteria) selected during the screening will be scrutinized in kinetic resolution and asymmetric reactions using resting cells and growing cells depending on the desired reaction. During this first approach, the reaction scale will be 20 mg of substrate and the reaction evaluation concerning conversion ande nantiomeric excess of the substrates and products will be performed on a GC and/or GC/MS equipped with chiral columns. Scale up of the reactions of interest will be the next step, by means of a 5L fermentor. Some microorganisms (Saccharomyces cerevisae, Serratia rubidea, Rhizopus oryzae, Aspergillus niger, Aspergillus oryzae, Rhodotorula glutinis, Corynebacterium aquaticum) of interest, already in use in our laboratories, are waiting scale up for lack of fermentor. Most selected substrates are low molecular weight epoxides, ketones, 1,2- and 1,3-diketones, non activated alkenes and acetates, of little commercial value. The biorcations will therefore aggregate chirality to the substrates thus producing valuable fine chemicals, useful as intermediates in the pharmaceutical and agrochemical industries. Isolation and evaluation of microorganisms of restricted and extreme ambients will be considered during the project development. The isolation of enzymes of interest will be the theme of future work. Finally, the collaboration with microbiologists and biochemists is essential to fully develop this project that has much interface between chemistry and biology. (AU)
