Studies toward the application of biocatalysis in the synthesis of bioactive molecules

Abstract

This research proposal aims to investigate applications of biocatalyzed reactions in the syntheses of molecules with biological activity.The main subject of investigation of this proposal is the reaction of enzymatic kinetic resolution of secondary alcohols and the reaction of ketone bioreductions. These reactions can be mediated by isolated enzyme or whole-cell of microorganism. We intend to investigate both strategies in the biocatalyzed reactions. The use of alternatives solvents, as ionic liquids, will also be investigated mainly to find an environmentally benign solvent or to find a more efficient solvent than water for the whole-cell reactions.The biocatalysis is an excellent methodology to reach high enantiomeric excesses of asymmetric molecules, besides it is an environmentally safe synthetic strategy, once many principles of green chemistry are followed, such as energy economy, low toxicity, possibility to use water or other safe solvents, as ionic liquids and scCO2, besides the recovery of the catalyst. The target molecules of this project, as flossonol, (+)-xilarenone and substituted aryltetralones, were isolated from natural sources and presented, respectively, anticancer, antioxidant and antimalarial activities. Using the biocatalyzed reactions as the key step in the proposed syntheses, these molecules and/or its analogues would be first synthesized. (AU)
