Abstract

Molecular diagnostics methods have been widely used to detect and quantify bacterial species from oral cavity. The long-term storage of bacterial DNA samples may compromise the genetic material of the species interfering in the final results providing by these methods. The aim of this study is to evaluate the effect of temperature and storage time on the final quality of the DNA samples from oral cavity. Clinical samples of saliva and supra-gingival biofilm will be harvested from 10 healthy subjects and assessed by DNA Checkerboard hybridization technique. Samples will be tested immediately after taken (baseline-control) and after 2 weeks, 1 month, 6 months or 12 months of storage at room temperature, 4°C and -20°C. In addition to the detection and counting of the bacterial species, a correlation between the samples from the proposed protocols will be performed. (AU)
