Expression of utrophin, CD68 and CD163 in skeletal muscle of dogs affected by progressive muscular dystrophy

Abstract

The muscular dystrophies are hereditary diseases with progressive and degenerative development of skeletal muscle. Duchenne Muscular Dystrophy (DMD) is the most common and most devastating in the forms of hereditary muscular dystrophy in men. DMD in humans is recessive X chromosome caused by mutations in the gene dystrophin. The DMD is caused by lack of dystrophin, a protein that binds to the cytoskeletal matrix extracellular and leads to muscle wasting and progressive weakness. The dog Golden retriever (GR) is used as an experimental model to study the Duchenne muscular dystrophy (DMD), because the disease is phenotypically and genotypically similar to this one. This animal model is ideal for similarity of the manifestations and their genotypic and phenotypic disease are called the Golden retriever Muscular Dystrophy (DMGR). In addition, dogs have body mass and muscle changes similar to those DMD patients. Due to the increase of crosses between animals various strains and knowing the possibility of spontaneous mutations that can lead to muscular dystrophy, it's fundamental importance to study this disease in dogs for better understanding in human patients. In literature, data on changes in muscle relatedo to muscular dystrophy in animal models are scarce. With that the present work, using dogs as Golden retrievers animal model for muscular dystrophy, intends to conduct an immuno- histochemical features that muscular injuries and establish comparison between these lesions and the alternation of the degenerative and regenerative processes . The aim of this study will be to study the expression of utrophin, CD63 and CD168 in diaphragm muscles, masseter, triceps brachial and femoral biceps muscle in the degenerative processes skeletal muscle affected dogs of different ages, methods for immunohistochemical analysis.
