Oxidative stress markers after cycles of adjuvant chemotherapy for colon cancer: effect of zinc supplementation

Abstract

To determine the oxidative stress after cycles of chemotherapy in patients with adjuvant therapy after surgery for colon cancer, with or without zinc supplementation. Subjects: Thirty adults (older than 18 years) from both genders will participate in this study. All patients underwent stage III colon cancer surgical resection and are starting adjuvant chemotherapy with capecitabine and oxaliplatin in HCFMRP- USP. Patients will be randomized into two groups. The first one (QT-Zn Group, n=15) will receive 70 mg/d of zinc orally for 12 weeks and the second one will receive placebo (QT-Placebo Group, n=15). The study will also include 30 healthy volunteers matched for age, gender and socioeconomic status, who will receive 70 mg /d of zinc supplement (C-Zn Group, n=15) or placebo (C-Placebo Group, n=15) for 12 weeks. Methods: The questionnaires about dietary intake (semiquantitative food frequency and food record), fatigue (FACIT-F) and questionnaires that assess the side effects of chemotherapy (CTCAE) will be evaluated. Anthropometry and bioelectrical impedance measurements will be made. Blood collection will be performed before the 1st, 2nd, 3rd and 4th cycles of chemotherapy (median duration of 21 days among cicles). The nutrients involved in oxidative stress (vitamin C and E, zinc and selenium) and markers anti and pro-oxidants (MDA, SOD, GPx and 8-isoprostane) will be determined. The control group will undergo the same procedures, except for chemotherapy. Statistical analysis will be made by the software Statistica 7.0, using ANOVA F-test or by Kruskal - Wallis, according to normality of variables. (AU)
