Abstract
Systemic diseases have been associated with episodes of hypertension, such as diabetes, hyperthyroidism, dyslipidemia and osteoporosis. The purpose of this study is to evaluate the development potential of the osteoblastic phenotype of mesenchymal stem cells derived from bone marrow of normotensive and hypertensive rats in the presence or absence of an inflammatory process. For this we use the colorimetric MTT assay (3 - [4,5-dimethylthiazol-2-yl] -2.5 - diphenyl tetrazolium bromide), measurement of total protein, measurement of alkaline phosphatase, detection and quantification of extracellular matrix, elisaimunoensaio, and Real-Time PCR (Polymerase Chain Reactions), and immunohistochemical analysis. Will be used 20 rats divided into 2 groups: normotensive and spontaneously hypertensive (SHR), and these will be subdivided into two other subgroups: presence or absence of periodontal disease. The animals will be euthanized 15 days after the ligation, and this time will be made to collect the bone marrow of the femurs of the animals. Stem cells are deposited in the appropriate culture medium until reaching confluence. The regions undergoing periodontal bandage will be removed for laboratory processing and paraffin embedding. Some series of sections will be stained with hematoxylin and eosin to perform histomorphometric analysis. The analysis will be performed through optical microscopy bright field, and the data will be analyzed statistically. Photographs of the slides are captured with the aid of a camera attached to a stereomicroscope. (AU)
