Effect of chronic ethanol consumption on the retinoic acid metabolism in the dorsal and lateral lobe prostate of the UCH rat

Abstract

Recent research has linked chronic ethanol consumption with the risk of prostate cancer (PCa). Retinoic acid (RA) is important for maintaining homeostasis in the adult prostate. Ethanol alters the RA metabolism and its concentration in many tissues, and thus, may contribute to the development of pathologies such as cancer. The chronic ethanol consumption increases the AR concentration in the prostate. Additionally, changes in concentration and on metabolism of RA also occur during PCa and benign prostatic hyperplasia, suggesting that the AR pathway is an important way for the development of pathologies in this gland. Moreover, AR can modulate the activity of matrix metalloproteinases (MMPs), which contribute to the initial and final stages of PCa. The mechanism by which chronic ethanol consumption alters the concentration of AR in the prostate and its influence on the activity of MMPs is not known and could contribute to research investigating the relationship between the chronic ethanol consumption with the development of PCa, besides bring new insights on dysfunctions ethanol-induced in this gland. Thus, this study aims to analyze the effects of low consumption (UChA rats) and high (UChB rats) of ethanol on proteins involved in the synthesis and metabolism of RA and on the enzymatic activity of MMPs on the dorsal and lateral lobes of the rat prostate. Will be assessed the expression and localization of the proteins ALDH1A1, ALDH1A2, ALDH1A3, CYP26A1, CYP26B1, CYP2E1 and CRABP, and the metabolic activity of the enzymes CYP26A1, CYP26B1, CYP2E1, MMP-2 and MMP-9. This study will provide new insights into the research area developed at the Department of Anatomy (IBB / UNESP) by the research team in Biology of Reproduction. (AU)
